Investigation of the maximum feasible volume of subretinal injections into rat and cynomolgus monkey eyes

Subretinal injection is widely used in gene therapy research on age-related macular degeneration and retinitis pigmentosa. We investigated a method of the maximum feasible volume for subretinal injections rats monkeys to support future studies evaluation new therapies these animals. Physiological saline was injected subretinally into eyes male adult (n = 3 eyes/group) cynomolgus eyes/group). The were then examined by ophthalmoscopy (slit lamp ocular fundus examinations), optical coherence tomography (OCT), intraocular pressure (IOP) measurements, or electroretinography (ERG). Rats received physiological at 1, 2, 5 µL/eye (2 sec/eye, bolus) (5 µL/min). Monkeys 50, 100, 150, 200, 250 (10 µL/sec). In all monkeys, successful visually confirmed bleb formation using OCT appropriate sites immediately after injection. IOP increased more than 2-fold µL/2 sec) compared with before but remained virtually unchanged other volumes. that 200 showed marked increase injection, functional abnormality observed ERG. conclusion, OCT, demonstrating rat monkey eyes. When solution injected, considered be µL/min) 150 µL/sec) based ERG results.
